Cílem projektu je uskutečnit expresní analýzu šesti cytotoxických derivátů kyseliny betulinové na nádorové linii CEM. Expresní profily budou porovnány s profily sandardních léčiv, diferenční data významně přispějí k pochopení mechanismu účinku a povedou k další validaci terapeutických cílů. Czech Republic is a country with a relatively high rate of cancer. The unsatisfactory results of anticancer therapy to date and the narrow spectrum of targeted drugs require active search for compounds with novel mechanisms of antitumour action. Our group is studying the mechanisms by which the triterpenoid compounds exert their cytotoxic effects. The most potent of these compounds induced apoptosis in a concentration less than IC50=0,2mM. We have identified cytochrome c as one unique target of b-keto betulinic acid. However, other potent compounds reveal a different kinetic of apoptosis induction with shifts in cell cycle distribution. Owing to the wide spectrum of possible targets we plan to use expression fingerprinting (profiling of mRNA/proteins) of a treated CEM cancer cell line. The expression profiles obtained will be compared to the profiles of drugs with identified mechanisms of action and further statistically analyzed. The differential data is expected to contribute to our understanding of the mechanism of action and facilitate target validation of triterpenoid compounds.
