Cílem projektu je studovat vývoj rostliny, expresi genů metabolismu cytokininů (CK) a hladiny CK a auxinů po aplikaci derivátů CK, testovat nová analoga CK a inhibitory biosyntézy, ověřit alternativní mechanismus deaktivace CK a zkoumat fyziologickou funkci nesekretované cytokinindehydrogenasy. Synthetic derivatives of plant hormones cytokinins tested so far in our laboratory affect cytokinin degradation by cytokinin dehydrogenase and/or cytokinin perception by receptors in vitro. The objective of proposed project is screening of developmental changes after application of selected cytokinin analogues to plants which will be accompanied by detailed study of expression of genes involved in cytokinin biosynthesis, perception and deactivation as well as by monitoring level of cytokinin metabolites and auxins. Newly synthesized cytokinin analogues and compounds acting as inhibitors of cytokinin biosynthesis will be also tested. In addition, an alternative mechanism of cytokinin deactivation by xanthine dehydrogenase will be verified and physiological function of non-secreted cytokinin dehydrogenase AtCKX7 will be elucidated. Developmental changes triggered by different compounds can be used to study distinct metabolic pathways involved in cytokinin homeostasis and the compounds may find an application for improvement of yield attributes of agricultural crops.
