Cílem navrhovaného projektu je vývoj nové, obecně použitelné metody pro detekci a stanovení množství stafylokokových enterotoxinů a zjištění faktorů, které jejich tvorbu ovlivňují. Dalším cílem je najít rychlou, spolehlivou a levnou metodu pro detekci aflatoxinogenních plísní. 19 staphylococcal enterotoxins (SE) have been described, but only 5 of them can be detected in foodstuffs by ELISA or RPLA. In recent years information about illness caused by these new enterotoxins especially by enterotoxin H is on the rise. However detection of these enterotoxins in foodstuffs is not carried out routinely. Project will predominantly apply electrophoresis and western blot. However, these methods do not allow the precise quantification that is required nowadays. Our research will be focused on investigation of the conditions under which S. aureus is able to produce SE, but also on their amounts. It is intended to develop a method for determination of SE based on mass detection. Parallel research activity will deal with investigation of toxins produced by fungi Fusarium and Aspergillus. The toxins will be isolated and determined quantitatively by HPLC. The amount of toxin will be correlated with the influence of relevant factors. The electrochemical method of detection of DNA and the specific part of gene that encodes production of aflatoxin B1 will be developed.
