Description
| - Předmětem studia budou embryonální kmenové a normální a maligní hematopoetické kmenové buňky a identifikace specifických molekulárních mechanismů společných pro uvedené typy kmenových buněk. Analýzy budou prováděné na několika úrovních zahrnujících detailní studium genové exprese na úrovni mRNA i proteinů, studium mechanismů a funkce signálních drah a studium biologických vlastností kmenových buněk in vitro a in vivo. Vybrané, dobře charakterizované a jednoduše testovatelné společné mechanismy budou použity k tvorbě účinných modelů, zejména s využitím lidských embryonálních kmenových buněk a myších modelů, na kterých budou probíhat komplexní funkční analýzy s cílem geneticky nebo chemicky ovlivnit výše uvedené mechanismy. Konečným cílem výzkumného záměru je nalezení molekul, nebo molekulárních regulačních drah představujících potenciální cíle pro vývoj a testování nových protinádorových terapeutik, případně komplexnějších terapeutických strategií.
- The goal of this project will be to profit from availability of unique cellular resources - mouse and human embryonic stem cells and normal and malignant hematopoietic stem/progenitor cells, to effectively employ a wide variety of current and complementary methodologies available to the consortium, and to exploit a proven/certified expertise in working. All this together will represent a powerful experimental platform aimed at addressing a nature of molecules and molecular mechanisms underlaying phenomena that are common to both normal and cancer stem cells. Integral part of this general goal is to implement multifaceted analytical approach that will cover studies of gene expression at both mRNA and protein levels, evaluating function of extra and intracellular signaling pathways. Expected outcome will be not only understanding but also a definition of molecules and/or molecular pathways to be tested as therapeutic targets, and cellular models that will make such testing feasible.
