Description
  • The aim of this paper is a sensitive HPLC-FD method that has been developed for quantification of ultra-trace amounts of OTA in foodstuffs of plant and animal origin. Food samples were cleaned using commercial immunoaffinity columns and analyzed by means of the HPLC method with fluorescence detection. The analytical column 5um x 150mm x 4.6mm with guard cartridge 5um x 4mm x 10mm was used at a temperature of 40 C. Acetonitrile-water-acetic acid (50:49:1 v/v/v) was used as a mobile phase with a flow rate 1 mL/min. Injection volume was 100 uL. Parameters of the fluorescence detector were Ex = 333 nm, and Em = 443 nm, respectively. Recoveries were 80-88% in the range 0.01 - 0.30 ug/kg according to type of sample. The average relative standard deviation of repeatability (RSDr) was 3.75%. Limit of quantification of the method (LOQ) ranged between 0.01 - 0.30 ug/kg depending on the type of sample matrix. Quality of laboratory results was provided by successful participation in international inter-laboratory proficiency testing (FAPAS, FERA, UK).
