Description
| - Polyaniline is the polymer of wide application potential but data on its biocompatibility are not plentiful and current study is hence focused on the determination of cytotoxicity, irritation and delayed-type hypersensitivity of polyaniline powders. Though the combination of the above methods is primarily intended for medical devices coming in contact with intact skin, mucosal membrane and breached or compromised skin surface for a prolonged period of time (24 to 30 days), data on cytotoxicity, which is one of the essential tests, can provide valuable information for further potential polyaniline applications in more advanced medical devices. To test the cytotoxicity effect of polyaniline samples, two cell lines were used, namely Human immortalized non-tumorigenic keratinocyte cell line and Human hepatocellular carcinoma cell line. The test was performed according to modified EN ISO 10993-5 (Biological evaluation of medical devices – Part 5: Tests for in vitro cytotoxicity). Skin irritation potential of polyaniline extracts was investigated by performing an in vivo patch test in 30 volunteers. Sensitization was studied on 25 non-pregnant healthy female of Dunkin-Hartley guinea-pigs. The skin irritation test and test for sensitization were performed according to EN ISO 10993-10 (Biological evaluation of medical devices – Part 10: Test for irritation and sensitization) at National Reference Center for Cosmetics and National Laboratory for Experimental Immunotoxicology, National Institute of Public Health, Prague, Czech Republic. The results from the above mentioned studies show that tested polyaniline does initiate neither skin irritation nor senzibilization. The cytotoxicity testing revealed different level of toxicity of samples which was correlated to their composition and preparation procedure.
