  • Cílem práce bylo monitorování a studium bakteriálních druhů, rozkládajících xanthan a gellan, ve vzorcích půd a odpadních vod. Pro odhady počtu degradačních bakterií byla použita metoda Most Probable Number, v níž posouzení rozkladu substrátu bylo provedeno metodou úbytku organického uhlíku. Kultury, které byly odpovědné za rozklad obou polysacharidů, byly u vybraných vzorků pasážovány a posléze izolovány. Výsledky ukázaly, že ve vzorcích odpadních vod se vyskytují bakterie degradující oba typy polysacharidů, v řádových množstvích 103 až 104 buněk / ml kalu, zatímco ve vzorcích půd byly výsledky poněkud rozdílné v závislosti na charakteru a typu zeminy.   • The main aim of the work became a monitoring of the xanthan- and gellan-degrading bacteria in soil and activated sludge samples and their study. Most Probable Number method was used for the estimation of counts of degrading bacteria. Their presences in the samples were proved by xanthan or gellan utilization in a liquid media. In some samples bacterial consortia responsible for the polysaccharide degradation were enriched. Results showed that bacteria degrading both polysaccharides were commonly present in most soil and sludge sample; their counts in sludge sample attained from 103 to 104 cells/ml while their soil numbers varied according to soil type.
