  • Adenozinovyjj receptor tipa A2B ehkspressiruetsja mnogimi immunnymi kletkami i oposreduet raz- vitie i podderzhanie khronicheskogo vospalenija pri bronkhial'nojj astme. V svjazi s ehtim adenozinovyjj receptor tipa A2B rassmatrivaetsja kak mishen' v terapii bol'nykh s nekotorymi zabolevanijami leg- kikh. Dlja analiza osobennostejj vzaimodejjstvija molekuly dannogo receptora s molekulami razlichnykh aktivatorov i ingibitorov primenjalos' komp'juternoe modelirovanie. Sozdany modeli adenozinovogo receptora tipa A2B i dvukh ego mutantnykh form (programma Modeller), a takzhe 9 aktivatorov i 15 ingibitorov dannogo receptora (programma ChemSketch). Na osnovanii poluchennykh modelejj proiz- veden raschet ligand-receptornykh vzaimodejjstvijj s ispol'zovaniem programmy AutoDock 4.0. Ustanov- lena predraspolozhennost' vzaimodejjstvija aktivatorov i ingibitorov s opredelennymi aminokislotami ligand-svjazyvajushhego centra adenozinovogo receptora tipa A2B.
  • Adenosine receptor A2B is a weakly studied target for bronchial asthma, chronic obstructive pulmonary disease therapy. For a structure-based design project, the three-dimensional information of the designated target is needed. Unfortunately, experimental structure data for A2B adenosine receptor and is not yet available. Homology building (program Modeller) was used to generate three-dimensional structure data using structure information of a homologous protein - A2A adenosine receptor. Then, three-dimensional structures of adenosine receptor ligands (9activators and 15inhibitors) were created using program ChemSketch. Activators and inhibitors were then docked into the active site of A2B adenosine receptor using the program AutoDock 4.0. Our results show several amino acids plays the most critical role in the binding of most activators and inhibitors. (en)
