Description
  • The aim of work was detection of micro-eukaryota in faecal samples from healthy people and people suffering from celiac disease and comparison of the results. DNA was isolated from faecal samples of 20 people. It was analyzed using the PCR-DGGE method focused on 18S rRNA genes and PCR focused on fungal ITS regions, both followed by sequencing, and it was tested for presence of 4 bacterial genera (Aspergillus, Candida, Saccharomyces and Penicillium) using real-time PCR. Sequences were compared with database of nucleotide sequences for generic designation. DGGE profiles showed high diversity and low inter individual similarity of the intestinal micro-eukaryota. The sequence analysis revealed presence of Blastocystis sp., Candida albicans and Malassezia restricta, each in one subject. Most of the sequences were similar to different nonspecified sequences from human gut metagenome DNA. Real time PCR detected Saccharomyces cerevisiae in five subjects. No significant differences were found between microflora of healthy and celiac people.
