  • Prokázali jsme vysoce afinitní vazbu proteinu HMGB1 a nádorově supresorového proteinu p53 k nové struktuře DNA, která tvoří smyčku DNA stabilizovanou hemikatenátem (zkratka hcDNA). Afinita p53 k hcDNA je 40-krát vyšší (Kd ~ 0.5 nM) než k přirozeným vazebným místům tohoto proteinu v genech (Mdm2 promotor). Vazbu p53 s hcDNA lze detegovat i pří desetinásobném hmotnostním přebytku kompetitorové DNA. p53 má vyšší afinitu k hcDNA než k minikružnicím DNA (bez funkčních specifických sekvencí pro vazbu p53) o stejné velikosti jako je velikost smyčky DNA v hcDNA.Tyto výsledky naznačují, že vysoká afinita p53 k hcDNA je pravděpodobně důsledek vazby tohoto proteinu k hemikatenátu DNA. Stabilní ternární komplexy hcDNA a HMGB1 byly prokázány pouze s intaktním p53. (cs)
  • We have observed that chromatin architectural protein HMGB1 and tumor suppressor protein p53 could bind with high affinity to a novel DNA structure that forms a DNA loop maintained at its base by a hemicatenane (termed hcDNA). The affinity of p53 for hcDNA is 40-fold higher (Kd ~ 0.5 nM) than for its natural specific binding sites within its target genes (Mdm2 promoter). Binding of p53 to hcDNA remains detectable in the presence of up to ~4 orders of magnitude of mass excess of competitor linear DNA, suggesting a high specificity of the interaction. p53 displays a higher affinity for hcDNA than for DNA minicircles (lacking functional p53-specific binding sequence) with a size similar to that of the loop within the hcDNA, indicating that the extreme affinity of p53 for hcDNA is likely due to binding of the protein to the hemicatenane. Only the full-length p53 can form stable ternary complexes with hcDNA and HMGB1.
