  • Jedním z významných problémů úpravy povrchové vody je sezónní masový rozvoj fytoplanktonu a s ním spojené zvýšené koncentrace AOM. Cílem této práce je charakterizovat AOM produkované sinicemi Anabaena flos-aqua a Microcystis aeruginosa a zelenou řasou Scenedesmus quadricauda pomocí relativní molekulové hmotnosti, popsat rozdíl ve složení mezi EOM a IOM a demonstrovat afinitu AOM k hlinitým a železitým destabilizačním činidlům. Z výsledků provedených analýz je patrný značný rozdíl v charakteru a množství proteinů obsažených jako součást EOM v porovnání s IOM. Rozdíl v produkci proteinů je také patrný v závislosti na druhu zkoumaného mikroorganismů a vývojové fázi kultury, kdy dochází k postupnému zvyšování podílu proteinů na celkovém množství AOM. Největší podíl proteinů na celkovém množství AOM produkovala kultura sinice M. aeruginosa, u které byly např. jako součást IOM identifikovány proteiny o relativní molekulové hmotnosti 21, 85, 234, 395, 470 kDa a nad 900 kDa.   • This paper aims at the characterization of algae organic matter (AOM) produced by the cyanobacteria Anabaena flos-aqua and Microcystis aeruginosa and the green algae Scenedesmus quadricauda. Further, it is focused on the description of differences in the composition of extracellular organic matter (EOM) and intracellular organic matter (IOM), and on the demonstration of AOM affinity to aluminum and iron destabilization reagents. The results from the conducted analyses imply a significant difference in the amount and properties of the proteins contained in EOM in comparison to IOM. The differences in the production of proteins also depend on the species of microorganism observed and its growth phase; ageing of the culture is accompanied by a gradual increase of the portion of proteins forming AOM. Using affinity chromatography, the proteins with relative mass weight around 60 kDa were isolated as a component of AOM of cyanobacteria A. flos-aqua and M. aeruginosa.
