Description
| - Prasačí cirkovírus 2 (PCV-2) patrí medzi malé, neobalené vírusy s ikozahedrálnou symetriou kapsidy. Jeho genóm je tvorený jednovláknitou cirkulárnou DNA. Vírus PCV-2 je asociovaný s ekonomicky významnými ochoreniami prasiat na celom svete, medzi ktoré patria napr. syndróm poodstavového multisystémového chradnutia odstavčiat, syndróm dermatitídy a nefropatie prasiat a taktiež sa podieľa na vzniku reprodukčných problémov prasiat. Užitočným diagnostickým nástrojom na monitorovanie replikácie vírusu v rôznych tkanivách sú monoklonálne protilátky proti kapsidovému proteínu vírusu PCV-2, ktorý je kódovaný génom v oblasti ORF2. Rekombinantné DNA technológie a in vitro selekčné techniky umožňujú produkciu rekombinantných monoklonálnych protilátok. Fágový displej a následná produkcia imunoglobulinových fragmentov v expresnom systéme E. coli je veľmi rozšírenou metódou prípravy scFv (single chain variable fragment) protilátok. Pričom rekombinantné scFv protilátky sa dajú použiť v rovnakých aplikáciach ako kon (cs)
- Porcine circovirus 2 (PCV2) is considered to be an important pathogen associated with a number of different syndromes and diseases in pigs. One of these syndromes - postweaning multisystemic wasting syndrome (PMWS) was initially recognized in weaning piglets of a high-health-status Canadian herd in 1991. Since then, PMWS has become an economically important disease in virtually all regions of the world that produce pigs. Because of strong immunoreactivity the capsid protein is the preferred antigen in variety of serological tests (Mahé et al., 2000). Testing for PCV2 antibodies in sera is routinely performed by indirect immunoperoxidase or immunofluorescence staining of PCV2-infected cell cultures (Ouardani et al., 1999). However, these tests require the cultivation of PCV2 virus on porcine cells, a procedure which can be very time-consuming and labor-intensive because of very slow viral replication cycle.
