Description
  • Využitie mikrosatelitov v overovaní parentity je už bežnou metódou. Od polovice roku 2001 je v Laboratóriu aplikovanej molekulárnej genetiky (LAMGen) na MZLU v Brne zavedená technika analýzy mikrosatelitných lokusov (MS) DNA na overovanie rodičovstva (parentity) u ošípaných, HD a koní. Súčasné výhody metód DNA sekvenovania, počítačových programov a rozvoj bioinformatiky uľahčil identifikáciu jednotlivých SNP markerov v genómovej DNA, ktoré postupne začínajú nahrádzať dnes využívané MS markery. Cieľom tohoto príspevku je informovať o našich výsledkoch využitia MS (stanovenie frekvencií najvýznamnejších mikrosatelitných markerov) a zhodnotenie súčasných a perspektívnych metód v overovaní parentity u HD.
  • Although the highly informative, multi-allelic microsatellite markers (MS) have dominated for now over ten years the field of molecular genetic studies in human and animals, either used as model organisms or studied for their interest in agriculture, an increasing importance is now given to single nucleotide polymorphisms (SNPs), that are merely di-allelic base substitutions of lower heterozygosity. However, contrariwise to microsatellites, for which the genotyping technology has only evolved from manual scoring of alleles, seen as PCR products of varying sizes on acrylamide gels, to a semiautomated method using sequencing mashines, the situation is not so clear concerning the genotyping of SNPs, for which a broad range or methodologies are available. In cattle, the challenge has been to identify a minimal set of SNPs with sufficient power for use in a variety of popular breeds and crossbred populations. (en)
