Description
  • The genus Armillaria causes serious demage to forest stands in all Europe. In the Czech republic six species are presented, each of them differs in pathogenicity. In forest nurseries, however, no study aimed at armillaria were performed. Because armillaria do not fructificate at this age of trees, isolation of DNA from soil were chosen as the most suitable method. Our study took place at two different levels, as both bare-root seedlings and containers were tested for fungal DNA. The Armillaria specific primer pair (AR1, AR2) based on conserved sequences within the so-called ITS region (ribosomal DNA) was used for direct amplification of DNA from soil samples by nested PCR. The individual species were distinguished by RFLPs analysis with restriction endonuclease Hinf I.
