  • Práce se zabývá využitím FT NIR spektrometrie ke stanovení čistých bílkovin (ČB) a kaseinu (K) v mléce kravském, kozím, ovčím a v ovčím mlezivu. Měření bylo prováděno na přístroji FT NIR Nicolet Antaris v rozsahu vlnových délek od 4000 do 10 000 cm-1 se 100 scany a rozlišením 8. Vzorky mléka byly před měřením zahřáté na 40 °C, protřepány a ochlazeny na 20 °C. Spektra vzorků byla měřena na integrační sféře v režimu reflektance (technika měřící absorpci záření po odrazu paprsku od povrchu vzorku, který byl umístěn v Petriho misce a měřící prostor vymezen transflektanční kyvetou). Každý vzorek byl proměřen třikrát a pro kalibraci bylo použito průměrné spektrum. Kalibrace byla vyhotovena pomocí PLS metody. Bylo dosaženo vysokých korelačních koeficientů kalibrace: kravské mléko ČB 0,943; K 0,964, ovčí mléko ČB 0,997; K 0,977; kozí mléko ČB 0,989, K 0.890, ovčí mlezivo ČB 0,983. Kalibrace byly ověřeny křížovou validací. Výsledky validačních korelačních koeficientů: kravské mléko ČB 0,923; K 0,910, ovčí mlék (cs)
  • Our work deals with a possibility of determination of true protein and casein in cow's, ewe's and goat's milk and in ewe's colostrums by FT NIR spectroscopy. Samples of milk were analysed by FT NIR in reflectance mode with the transflectance cuvette. The values of correlation coefficient of calibration were as follows: cow's protein 0.943; cow's casein 0.964; ewe's protein 0.997; ewe's casein 0.977; goat's protein 0.989; goat's casein 0.890; ewe's colostrums protein 0.983. Calibration was tested using the same set samples by cross validation method. The values of correlation coefficient of validation were as follows: cow's protein 0.923; cow's casein 0.910; ewe's protein 0.994; ewe's casein 0.963; goat's protein 0.972; goat's casein 0.814; ewe's colostrums protein 0.871. NIRs results were compared with reference data and no significant differences between them were found (P = 0.05).
