Description
  • Glutathion a metalothionein hrají v organizmu klíčovou roly v detoxifikaci organizmu před různými chemickými látkami. V této práci jsme studovali interakci glutathionu redukovaného a oxidovaného, nitrosoglutathionu a MT s platinovými cytostatiky (cisplatin, carboplatin and oxaliplatin). • Glutathione and metallothionein (MT) play a key role in detoxification of chemical compounds coming to an organism. Due to their high affinity to heavy metals, homeostatic control and detoxification of the metals are one of their main physiological functions. Objective. The aim of this work was to study and characterize interactions of glutathiones (reduced GSH, oxidized GSSG and S-nitrosoglutathione GSNO) and MT with platinum group metals and cytostatics (cisplatin, carboplatin and oxaliplatin). Materials and methods. We used different electrochemical techniques (cyclic voltammetry, differential pulse voltammetry and chronopotentiometric stripping analysis). Measurements were performed with AUTOLAB connected to VA-Stand 663, using standard cell with three electrodes. We observed that signal of GSH decreased with increasing concentration of a heavy metal. Moreover, presence of heavy metals catalysed decomposition of GSNO, thus, nitric oxide was released. Conclusion. MT could markedly decrease therap
