Description
  • Apical dominance is a dominance of the shoot apex over the axillary buds. It is well-known that auxin accumulated in the apex is transported towards the roots by polar transport. The mechanism, whereby polar auxin transport in the shoot regulates outgrowth of axillary buds, is unknown. In our experiments revealed Northern blot analyses PsDRM1 decrease after decapitation. If the apical bud in decapitated plants was substituted by IAA no change or even elevation occurred after 24 h. BA application to the inhibited bud led to disappearance of PsDRM1 after 24 h. These results suggesting that application of IAA and BA influenced the expression patterns of PsDRM1 coincidentally with their physiological influence on apical dominance. Homologous genes for auxin cell membrane transporters (PIN 1 and AUX 1) were cloned and sequenced in pea. PsAUX1 and PsPIN1 probes are used for studies of their role in apical dominance in pea.

  • Apikální dominance je dominancí lodyžního vrcholu nad axilárními pupeny. Je známo, že auxin akumulující se ve vrcholu je polárním transportem transportován ke kořenům. Mechanizmus, kterým polární transport auxinu v lodyze reguluje vyrůstání axilárních pupenů není známý. V našich experimentech ukázala hybridizace RNA snižování exprese PsDRM1 po dekapitaci. Pokud apikální pupen dekapitovaných rostlin byl nahrazen IAA, nedošlo k žádným změnám, nebo se ukázalo nevýrazné zvýšení po 24 hodin. Aplikace BA k inhibovaným pupenům vedla k vymizení PsDRM1 po 24 hodin. Tyto výsledky naznačují, že aplikace IAA a BA ovlivňuje expresi PsDRM1, což je v souladu s jejich rolí v apikální dominanci. Homologní geny auxinových membránových přenašečů (PIN1 a AUX1) byly klonovány a sekvenovány u hrachu. (cs)
