  • A set of PEMV and PSbMV isolates collected on pea plants in five locations in the Czech Republic (determined by symptoms on pea plants and confirmed by ELISA) was characterized by biological and molecular tools. The most severe isolates were selected for sequencing and the development of constructs for pea transformation. The strategy of induction resistance to viruses was based on pathogen-derived resistance (PDR) and posttranscriptional gene silencing (PTGS). The full length CP-PEMV cDNA was cloned in sense orientation (plasmid pWell105b) and 309 bp fragments of CP-PSbMV in sense and antisense orientation (plasmid pWell07a) and inserted into Agrobacterium tumefaciens strain EHA 105. Commercial cultivars of peas were transformed and T1 seed generations of GUS/PCR positive transgenic plants were obtained. Viral infection of the progeny of pea plants will be studied further.

  • Izoláty PEMV a PSbMV získané na pěti lokalitách České republiky byly charakterizovány pomocí biologických a molekulárních přístupů. Nejsilnější izoláty byly vybrány pro tvorbu konstruktů vhodných pro transformaci hrachu. Zvolená strategie byla založena na dvou možných mechanismech, na patogenem indukované rezistenci (PDR) a postranskripčním zhášení genů (PTGS). Kompletní cDNA CP-PEMV byla klonována v sense orientaci (plazmid pWell105b) a 309 bp dlouhé fragmenty CP-PSbMV v sense a antisense orientaci (plazmid pWell07a) a vloženy do Agrobacterium tumefaciens kmene EHA 105. Těmito byly transformovány komerční kultivary hrachu a následně bylo získáno T1 semenné potomstvo GUS/PCR pozitivních transgenních rostlin. Průběh virové infekce v transgenních rostlinách bude dále studován. (cs)
