  • Background DNA flow cytometry describes the use of flow cytometry for estimation of DNA quantity in cell nuclei. The method involves preparation of aqueous suspensions of intact nuclei whose DNA is stained using a DNA fluorochrome. The nuclei are classified according to their relative fluorescence intensity or DNA content.   • Kvalitní stanovení absolutního obsahu DNA u rostlin za pomocí průtokové cytometrie není triviálním úkolem. Ačkoli existuje několik ověřených protokolů a některé faktory kontrolující přesnost a reprodukovatelnost byly identifikovány, stále zůstávají některé problémy: (1) nezbytnost čerstvé tkáně komplikuje transfer %22doma připravených%22 vzorků a/nebo jejich skladování: (2) úloha cytosolických složek interferujících s kvantitativním DNA barvením není příliš známa: a (3) užití sady 4 mezinárodně uznaných DNA referenčních standardů zůstává stále nerealizovaným cílem.
