  • Bakteriální kmen Escherichia coli exprimující dva různé cytokininové receptory Arabidopsis thaliana, CRE1/AHK4 a AHK3, byl využit pro studium relativní citlivosti těchto receptorů k různým cytokininům. Oba receptory byly nejcitlivější k isoprenoidním cytokininům trans-zeatinu a isopentenyladeninu, avšak výrazně se lišily v rozpoznání ostatních cytokininů. CRE1/AHK4 rozeznával v 1 #M koncentraci pouze trans-zeatin, zatímco AHK3 rozeznával také cis-zeatin a dihydrozeatin, i když s nižší citlivostí. Podobně, CRE1/AHK4 nebyl aktivován ribosidy a ribotidy cytokininů, zatímco AHK3 ano. Srovnání s výsledky rostlinného biotestu, využívajícího semenáčky Arabidopsis s ARR5::GUS fúzním genem fungujícím jako cytokininový reportér, ukázalo podobnou relativní odezvu. Výjimkou byly cytokininy s aromatickým postranním řetězcem, které vykazovaly mnohem vyšší aktivity než v bakteriálním testu.   • Strains of Escherichia coli that express two different cytokinin receptors of Arabidopsis thaliana, CRE1/AHK4 and AHK3, were used to study the relative sensitivity of these receptors to different cytokinin compounds. CRE1/AHK4 recognized only trans-zeatin while AHK3 recognized also cis-zeatin and dihydrozeatin, although with a lower sensitivity. In addition, AHK3 was activated by cytokinin ribosides and ribotides, which did not activate CRE1/AHK4. Comparisons using the ARR5::GUS fusion gene as a cytokinin reporter in Arabidopsis showed similar relative degrees of responses in planta, with the exception that aromatic cytokinins showed much higher activities than in the bacterial assay. These results indicate that the different local concentration of the diverse cytokinin metabolites might have specific functions in the numerous cytokinin-regulated processes, which may depend in turn on different receptors and their associated signalling pathways.
