  • V této práci jsme stanovili účinky dehydroepiandrosteronu (DHEA) na intracelulární hladiny mRNA receptorů aktivovanými peroxisomálními proliferátory (PPAR) a PPAR-γ koaktivátorů v buněčné linii HepG2 za pomoci metody kvantitativní real-time RT-PCR. Ovlivnění 100 μmol/l DHEA po dobu 2-20 hodin vedlo ke zvýšení hladin sledovaných mRNA s časem. Po 20 hodinách jsme pozorovali zvýšení mRNA PPAR-α, -γ1 a ?γ2 i koaktivátorů PGC-1α a -1β na 157, 161, 155, 656 a 475 % (p < 0,05) oproti kontrole. Ovlivnění aktinomycinem D po dobu 2,5 ? 8 hodin odhalilo signifikantní stabilizační účinek DHEA na mRNA PPAR-γ1 a PGC-1α v obou časových intervalech. 8-hodinová inkubace s aktinomycinem D také vyvolala mírný, ale signifikantní stabilizační účinek na PGC-1β. Z výsledků vyplývá, že DHEA může řídit přeměnu těchto transkriptů. Po 20-hodinovém působeni cykloheximidu došlo k signifikantnímu poklesu bazálních hladin mRNA PPAR-α a PGC-1α, zatímco hladiny mRNA PPAR (cs)
  • To investigate the effect of dehydroepiandrosterone (DHEA) on intracellular mRNA levels of peroxisome proliferator-activated receptors (PPARs) and PPAR-gamma coactivators (PGCs), we conducted a quantitative real-time RT-PCR study using HepG2 cells. Treatment with 100 micromol/l DHEA for 2-20 h caused a time-dependent elevation of mRNA levels in the cells. Upon 20 h, PPAR-alpha, -gamma1, and -gamma2 mRNAs and PGC-1alpha and -1beta mRNAs increased to 157, 161, 155, 656, and 475% of control levels, respectively (p < 0.05 each). Treatment with actinomycin D for 2.5-8 h revealed a significant stabilization effect of DHEA on PPAR-gamma1 and PGC-1alpha mRNAs at both 2.5 and 8 h incubation periods and a mild but significant stabilization effect on PGC-1beta mRNA at the 8 h incubation period suggesting that DHEA can modulate turnover of these mRNA transcripts.
