| - Velmi malé změny mRNA stability mohou ovlivnit rozhodující body buněčných energetických drah. Cílem této studie bylo prozkoumat vliv energeticky bohatých substrátů na stálé hladiny mRNA receptorů aktivovaných proliferátory peroxisomů (PPARs) a PPAR-gamma koktivátorů (PGCs). Byla provedena kvantitativní RT-PCR studie pro vyhodnocení vlivu nulové nebo normální (5 mmol/l) glukózy a kyseliny olejové (0,3 mmol/l) na hladiny mRNA (PPARs) (PGCs) v hepatomových HepG2 buňkách. Hladiny mRNA. PGC-1alfa byly signifikantně zvýšené v buňkách ovlivněných nedostatkem glukózy (123% kontrolní hodnoty; p < 0,05), zatímco hladiny mRNA PGC 1beta byly signifikantně zvýšené v buňkách ovlivněných kyselinou olejovou (134% a 160% kontrolní hodnoty v buňkách ovlivněných nulovou glukózou s kyselinou olejovou a ovlivněných normální glukózou s kyselinou olejovou; p < 0,05) během 0,5 h inkubace. The aim of this study was to investigate the impact of energy abundant substrates on peroxisome proliferator-activated receptors (PPARs) and PPAR-. coactivators (PGCs) mRNA?s steady-state levels. Quantitative RT-PCR study was performed to assess the effect of zero or normal (5 mmol/l) glucose and/or oleic acid (0.3 mmol/l) on mRNA levels of (PPARs) (PGCs) in HepG2 cells. PGC-1alfa mRNA was significantly upregulated in glucose deprived cells (123 % of the control level; p < 0.05), while PGC-1ß mRNA was significantly enhanced in oleate-fed cells (134 % and 160 % of control levels for zero glucose plus oleate and normal glucose plus oleate, respectively; p < 0.05) during the 0.5 h incubation.
