| - The quarternary benzo[c]phenanthridine (QBA) alkaloid, sanguinarine (SG) is known for a range of biological activities (antimicrobial, anti-inflamatory, etc.). Only recently has reduction of SG been designated as the first step in its metabolism and detoxification, yielding dihydrosanguinarine (DHSG) as a first metabolite. The toxicity of DHSG has never been studied which is rather surprising seeing that DHSG is identified as the toxic component involved in the Epidemic dropsy syndrome, caused by Argemone mexicana oil. This syndrome related to oxidative stress. The aim of our study was to gather reliable data on any subchronic toxicity and oxidative stress related to (DHSG) in oral study in rats. Rats were fed a diet containing 100 ppm and 500 ppm of DHSG for 90 days. The daily dose of DHSG for 100 ppm and 500 ppm groups, respectively, fell from 17 and 80,8 mg/kg b.w. at the beginning to 7.83 and 35,5 mg/kg b.w., at the end of the experiment, i.e. averaged doses over 90 days were 12,4 and 58,2 mg/

- Kvarterní benzo[c]fenantridinový alkaloid (QBA), sanguinarin (SG) je znám pro své široké spektrum biologických aktivit (antimikrobiální, protizánětlivou, atd.). Jako první krok metabolizace a detoxifikace SG byla prokázána jeho redukce, při které vzniká dihydrosanguinarin (DHSG). Toxicita DHSG nebyla zatím nikdy studována i přesto, že se o DHSG mluví jako o toxické komponentě v Epidemic dropsy syndromu, způsobeném olejem z Argemone mexicana. Tento syndrom je spjat s oxidačním stresem. Cílem naší práce bylo získat spolehlivá data o subchronické toxicitě a oxidačním stresu souvisejícím s DHSG ve studii na potkanech. Potkani byli krmeni dietou obsahující 100 ppm a 500 ppm DHSG po dobu 90 dní. Denní dávka DHSG, pro skupiny obsahující 100 ppm a 500 ppm , byla 17 a 80,8 mg/kg váhy potkana na začátku experimentu a 7,83 a 35,5 mg/kg váhy potkana na konci experimentu. Průměrná spotřeba krmiva činila 12,4 a 58,2 mg/kg váhy potkana. Spotřeba krmiva a hmotnost potkanů byla sledována dvakrát týdně. V měsíčníc (cs)
