  • Silybin, the main flavonolignan from Silybum marianum, is well recognized as an antioxidant and free radiacal scavenger with the capacity for regenerating liver cells. The aim of the present contribution was to investigate silybin conjugate formation in primary cultures of rat and porcine hepatocytes. For silybin determination HPLC method with SPE was used with several glucuronides of silybin standards. The amount of total conjugated silybin was evaluated after enzymatic cleavage. Several hydrophylic metabolites of silybin, namely glucuronides and sulphates, were found. Prefered position of transformation was 7 and 20 of diastereomer A.

  • Silybin, hlavní flavonolignan Silybum marianum, je známý jako antioxidant a lapač volných radikálů se schopností regenerace jaterních buněk. Cílem práce bylo sledování produkce konjugátů silybinu v primárních kulturách potkaních a prasečích hepatocytů. Pro stanovení silybinu a jeho metabolitů byla použita HPLC metoda s extrakcí na pevnou fázi a několik standardů glukuronidů silybinu. Celkové množství konjugovaného silybinu bylo zjišťováno po enzymatickém štěpení. Bylo nalezeno několik hydrofilních derivátů, glukuronidů a sulfátů. Preferovaná pozice metabolizace diastereomeru A byla 7 a 20. (cs)
