  • Cytokininy regulují buněčné dělení a diferenciaci, jakož i řadu dalších vývojových procesů u rostlin. První stupeň biosyntézy cytokininů v rostlinách huseníčku (Arabidopsis thaliana L.) je katalyzován adenosinfosfátisopentenyl transferázami (AtIPT) lokalizovanými v plastidech nebo cytoplasmě. Mechanismy regulující aktivitu AtIPT a koncentraci cytokininů ve vztahu k homeostázi cytokininů nejsou dosud plně objasněny. Práce prokázala, že zvýšená exprese AtIPT3, tj. jednoho z členů rodiny AtIPT, zvyšuje produkci isoprenoidních cytokininů. AtIPT3 je rovněž substrátem farnesyltransferázy, tj. enzymu katalyzujícího farnesylaci proteinů. Farnesylace AtIPT3 směruje enzym do jádra či cytoplasmy, zatímco nefarnesylovaný protein je lokalizován v plastidech. Bylo zjištěno, že různá vnitrobuněčná lokalizace farnesylovaného a nefarnesylovaného proteinu úzce souvisí s biosyntézou různých typů cytokininů a že cystein-333 v molekule AtIPT3 je klíčový pro jak pro farnesylaci, tak katalytickou aktivitu AtIPT3. The first step of cytokinin biosynthesis in Arabidopsis (Arabidopsis thaliana) is catalyzed by adenosine phosphate-isopentenyl-transferases ( AtIPT). The enzymes are localized in plastids or the cytoplasm where they utilize the intermediate dimethylallyldiphosphate from the methylerythritolphosphate or mevalonic acid pathways. However, the regulatory mechanisms linking AtIPT activity and cytokinin biosynthesis with cytokinin homeostasis and isoprenoid synthesis are not well understood. Here, we demonstrate that expression of AtIPT3, one member of the adenosine AtIPT protein family in Arabidopsis, increased the production of specific isopentenyl-type cytokinins.
