Description
| - DNA repair associated with DNA replication is important for the conservation of genomic sequence information, whereas reconstitution of chromatin after replication sustains epigenetic information. We have isolated and characterized mutations in the BRU1 gene of Arabidopsis that suggest a novel link between these underlying maintenance mechanisms. Brul plants are highly sensitive to genotoxic stress and show stochastic release of transcriptional gene silencing. They also show increased intrachromosomal homologous recombination and constitutively activated expression of poly (ADP-ribose) polymerase-2 (AtPARP-2), the induction of which is associated with elevated DNA damage. Brul mutations affect the stability of heterochromatin organization but do not interfere with genome-wide DNA methylation. - Reparace DNA asociovaná s replikací DNA je důležitá pro zachování informace v geonomových sekvencích, zatímco rekonstituce chromatinu po replikaci zachovává epigenetickou informaci. Izolovali jsme a charakterizovali mutaci v genu BRU1 huseníčku, který ukazuje nová propojení těchto udržovacích mechanismů. Rostliny Bru1 jsou velmi citlivé ke genotoxickému stresu a vykazují stochastické uvolnění transkripčního tlumení genů. Vykazují zvýšenou intrachromosomální homologní rekombinaci a konstitutivně aktivují expresi poly (ADP-ribose) polymerasy 2 (AtPARP-2), jejíž indukce je spojena s poškozením DNA. Mutace Bru1 ovlivňuje stabilitu organizace heterochromatinu, ale neovlivňuje geonomovou methylaci DNA. BRU1 kóduje nově identifikovaný jaderný protein se dvěma předpovězenými doménami pro interakci protein-protein.
