Description
  • The reductively degradable multiblock polymer designed for use in drug delivery was synthesized by oxidative polycondensation of the PEG-cysteine derivatives containing amide bonds between PEG and cysteine and reductively degradable disulfide bonds between cysteine residues. Polymer degradation and release of doxorubicin linked to the carrier through acido-labile hydrazone bond were studied. The rate of amide bond hydrolysis was negligible compared with that of reduction of disulfide bonds. The hydrazone bond was stable at pH 7.4 but 68% of the drug was relased after 24 at pH 5.

  • Reduktivně štěpitelné multiblokové polymery byly navrženy jako polymerní nosiče léčiv. Byly připraveny oxidativní polykondenzací PEG-cysteinových derivátů, obsahujících mezi blokem PEG a cysteinem amidovou vazbu a reduktivně štěpitelné disulfidové můstky mezi dvěma cysteinovými deriváty. Byla studována jak degradace samotného polymerního nosiče, tak i uvolnění cytostatika doxorubicinu, připojeného k nosiči acidolabilní hydrazonovou vazbou, z polymerního nosiče. Bylo zjištěno, že rychlost hydrolýzy amidové vazby byla ve srovnání s rychlostí reduktivního štěpení S-S vazeb zanedbatelná. Zatímco v pufru pH 7,4 nedocházelo k hydrolýze hydrazonové vazby, kterou bylo léčivo připojeno k polymernímu nosiči, v pufru pH 5,0 bylo za 24 h uvolněno z polymerního konjugátu 68% navázaného léčiva.
