  • Byl navržen prostorově dvourozměrný matematický model zařízení pro imunoanalytické stanovení lidských IgG protilátek s použitím Proteinu A imobilizovaného na vnitřních stěnách mikrokanálu.Mikročip se skládá z centrálního kanálu a čtyř ramen pro uvedení a dávkování vzorku nebo pufrů. Konvektivní transport iontů je zajištěn elektroosmotickým mechanismem při vloženém vnějším elektrickém poli. Bylo zjištěno, že inkubační doba imunoanalytického stanovení může být redukována pouze na několik metod. Teoreticky zjištěná kalibrační křivka imunoanalýzy je v dobrém souladu s publikovanými experimentálními výsledky. (cs)
  • We have proposed a spatially two-dimensional model ofmicrochip for immunological determination of humanimmunoglobulin G (IgG) with the use of Protein A (PA)immobilized on the internal walls of a microchannel. Themicrochip consists of the central channel with theimmobilized antigen and four channels for theintroduction of electrolytes. The convection flow in themicrodevice is induced by the imposed difference ofelectric potential (electroosmosis).We have analyzed the incubation phase of theimmunoassay. The effects of the antibody concentrationin a sample, the imposed difference of electric potential,and the surface heterogeneities in the reaction zone on thesaturation time have been studied. We have found that theincubation period of the quantitative immunoassay can becarried out within few minutes in the proposedmicrodevice, which enables fast diagnostics. Thecomputed calibration curve of the immunoassay is in agood agreement with the recently published experimentalresults.
