  • Cílem toto práce bylo sledování adheze bakteriálních buněk na povrch několika nosičů metodou analýzy obrazu. Gram pozitivní bakterie Rhodococcus erythropolis byla kultivována v minimálním médiu s fenolem o koncentraci 0,3g/l jako jediným zdrojem uhlíku a energie v přítomnosti několika materiálů (nosič) s rozdílnou hydorfobitou. Hydrofobita nosiče byla stanovena kapkovou metodou. Buňky přisedlé na povrch pevného nosiče byly obarveny fluorescenční barvou Syto 13 a pozorovány pomocí fluorescnčního a konfokálního mikroskopu. Among them are: enumeration of sessile cells after detachment, colorimetric method after washing, staining and distaining of sessile cells with crystal violet or safranin, or microscopic observation (light, confocal or electron microscopy). Gram-positive bacteria Rhodococcus erythropolis was cultivated under different cultivation conditions in various systems (shaked flasks or modified Robbin?s device) in mineral medium. Phenol (0.3 or 0.7g/l) was used as sole carbon source. The ability of materials with different hydrophobicity and composition to attach cells was monitored. For this purpose the materials with wide hydrophobicity range were prepared (Department of Polymers, Institute of Chemical Technology Prague).
