  • The aim of this work was to study the physiological changes of immobilized brewing yeast Saccharomyces cerevisiae in a continuous reactor at three different temperatures (15, 20 and 25°C). During the continuous experiment the viability of free and immobilized cells was followed using vital staining and flow cytometry. The results of viability determination were used to calculate the specific death rate of the immobilized cells (Kd). Further, the flow cytometry was used to follow the glycogen content of cells and the number of budding cells.

  • Cílem této práce bylo studovat fyziologické změny imobilizovaných pivovarských kvasnic Saccharomyces cerevisiae v kontinuálním reaktoru pri různých teplotách kultivace (15, 20 a 25°C). V průb?hu kontinuálního pokusu se sledovala životaschopnost volných a imobilizovaných kvasinek pomocí fluorescenčního barviva a průtokové cytometrie. Výsledky byly použity ke stanovení specifické rychlosti odumírání imobilizovaných kvasnic (Kd). Průtoková cytometrie byla dále použita ke sledování obsahu buněčného glykogenu a ke stanovení počtu pučících buněk. (cs)
