  • The method by means synthetic substrate BAPNA (N--benzoyl-DL-arginin-p-nitroanilid) was used for trypsin-inhibitor activity (TIA) determination in pea seeds. To the extraction of proteins is used glycine buffer pH 11 containing urea and EDTA, 2 h stirring and centrifugation. Synthetic substrate BAPNA is hydrolysed by trypsin and the absorbance of dislodged yellow p-nitroanilid is measured at 410 nm. The changes of TIA during germination and the following microwave treatment and the conventional drying of germinated pea to the final moisture 12 - 14 % were evaluated.

  • Ke stanovení aktivity trypsin inhibitoru (TIA) byla použita metoda se syntetickým substrátem BAPNA (N--benzoyl-DL-arginin-p-nitroanilid). K extrakci bílkovin byl použit glycinový pufr pH 11 obsahující močovinu a EDTA, 2 h míchání a odstřeďování. Syntetický substrát BAPNA je hydrolyzován trypsinem a měří se absorbance žlutého p-nitroanilidu při 410 nm. Byly vyhodnoceny změny TIA během klíčení a následujícího mikrovlnného ohřevu a při konvenčním sušení naklíčeného hrachu do sušiny 12 - 14 %. (cs)
