| - In the present study we investigated the role of NO-dependent events in meiotic maturation in pig oocytes. It has been suggested that NO-NOS has important physiological roles in a variety of reproductive processes such as follicular development, ovulation, and spermatogenesis. The synthesis of NO is catalyzed by three distinct isoforms of NOS: neuronal (nNOS), endothelial NO (eNOS) and inducible NOS (iNOS). Of the three NOS isoenzymes, the ovaries express eNOS and iNOS but not nNOS. In the present study, we investigated the role of iNOS on meiotic maturation of pig oocytes in vitro. Cumulus-oocyte complexes (COC) collected from porcine ovaries were cultured in a modified M199 medium with iNOS inhibitors L-canavanine. The addition of L-canavanine to the culture medium had a significant impact on meiotic development, and L-canavanine markedly inhibited the oocyte meiotic maturation. This iNOS inhibitor increased the percentage of atypical oocytes compared with the control group.
