  • The aim of our work was to enumerate, isolate and identify bifidobacteria in infant faeces. Thirty three fully breast-fed infants in the age between 1 and 12 weeks were investigated. FISH kit for the quantitative detection of Bifidobacterium sp. was compared with culture method. Bifidobacteria in faecal samples were enumerated and isolated using TPY agar modified by the addition of mupirocin (100 mg/L) and glacial acetic acid (1 ml/L). Pure cultures were identified to the genus level using fructose-6-phosphate phosphoketolase test, and to the species level using API 50 CHL and API ID 32 A Rapid tests followed by computer program Bacter. The identification to the species level was also carried out by PCR method using species-specific primers. Bifidobacteria counts in most faecal samples determined by both TPY agar and FISH procedure were from 108 to 1011 CFU/g. Three infants did not contain any bifidobacteria in their faecal samples.
