Description
| - Precision and reproducibility of FACS analyses carried out in the cuvette flow cell are affected by quality of the cuvette itself which might be significantly affected by contamination coming from samples. It can increase background fluorescence and cause light scattering, resulting in higher CV and lower precision. Using Hoechst 33342 and propidium iodide staining in our experiments on a BD FACSAria instrument, we encountered increasing Bright Bead %Robust CV (%rCV), especially in channels related to UV or violet lasers, and problems passing the CS&T procedure. Long-lasting cleaning procedures led only to temporary suppression of %rCVs and affected the lab workflow. We presumed that turbulent convection within the flow cell should produce more effective washing in comparison to linear flow. Here we present a simple and effective procedure that maintains the condition of the BD FACSAria cuvette flow cell and can be easily executed on a daily basis.
