Description
| - Imunitní odpověď k F. tularensis je převážně závislá na buněčné odpovědi. V poslední době se však ukazuje, že důležitou úlohu v časné fázi imunity hrají B buňky. My jsme chtěli zjistit, jestli kultivace bakterií s B buňkami je nějakým způsobem ovlivní. Pro studium jsme použili lidskou B buněčnou linii Ramos RA-1, myší B buněčnou linii A20 a primární kulturu myších splenocytů, které jsme infikovali F. tularensis LVS, F. tularensis FSC200 a zabitou bakterií FSC200 in vitro. Počet bakterií detekovaný pomocí CFU se za 24 hodin zvýšil o 1 logaritmus. Pomocí transmisní elektronové mikroskopie jsme detekovali za 24 přibližně 20% buněk infikovaných buněk. Inkubace lidské buněčné linie Ramos a myších slezinných CD19 buněk s F. tularensis vedla buňky k apoptóze. Přibližně 30% buněk bylo apoptických po 24 hodinách infekce a 70% po 48 hodinách infekce. Apoptóza byla potvrzena Western blotem použitím anti-PARP protilátky. Tato studie ukazuje na schopnost F. tularensis vnikat do B buněk a indukovat jejich apoptóz (cs)
- Immunity to Francisella tularensis is largely mediated by T lymphocytes but an important role of B lymphocytes in early stage of infection was previously uncovered. We wanted to find out if F. tularensis is able to infect B cells and/or influence them by direct contact. To investigate this possibility we infected B cell lines from mouse (A20) or humans (Ramos RA-1), or primary mouse spleen cells, with F. tularensis LVS and F. tularensis FSC200 in vitro. In all cases, we infetected bacteria on the cell surface and inside the B cells using transmission electron microscopy. More than 20% cells were infected by microbes after 24 h. The number of bacteria, determined by CFU, increased about 1 log during 24 h. Infection with live bacteria led to apoptosis of Ramos cells and mouse CD19 spleen cells. Approximately 30% of cells were apoptotic after 24 h and 70% after 48 h, independently of the F. tularensis strain, while only 10% of noninfected cell were apoptotic at either time point.
