| - The protoplasts of S. verrucosum PI 161173 were UV-irradiated to inactivate the nuclei after determining the optimal UV dose (germicidal lamp - 254 nm, 370 µW . cm-2 for 10 min). The cellular organelles of the S. tuberosum (cv. Karin, cv. Komtesa, cv. Kordoba) protoplasts were inactivated. Concentrations (all in mM) of iodoacetamide (IOA; 0.125; 0.25; 0.3; 0.4; 0.5; 1; 2; 5), iodoacetic acid (IOAA; 0.125; 0.2; 0.25) and rhodamine 6G (RH6G; 0.05; 0.1; 0.125; 0.25) were tested and optimal concentrations (0.4 mM IOA and 0.2 mM IOAA) were determined for use in asymmetric somatic hybridization. The UV irradiated protoplasts and the protoplasts with inactivated metabolism were then electrofused. Regeneration of the fused protoplasts and nonfused treated protoplasts (UV or 0.2 mM IOAA or 0.4 mM IOA) were evaluated. Fused protoplasts were more viable and prospered better than treated nonfused controls. Plant regeneration in combination ver (UV) + tbr cv. Karin (0.4 mM IOA) was recorded.

- Inaktivace buněčného jádra a organel byla studována u protoplastů S. verrucosum PI 161173 a S. tuberosum (cv. Karin, cv. Komtesa, cv. Kordoba). Protoplasty S. verrucosum byly ošetřeny UV-zářením (inaktivace jader). Byla stanovena optimální dávka UV záření (germicidní lampa s vlnovou délkou 254 nm, intenzitou záření 370 µW . cm-2 po dobu 10 min). Byla uskutečněna inaktivace semiautonomních organel u protoplastů S. tuberosum. Byly testovány různé koncentrace metabolických inhibitorů: jodoacetamid (IOA; 0,125 mM; 0,25 mM; 0,3 mM; 0,4 mM; 0,5 mM; 1 mM; 2 mM; 5 mM), jodooctová kyselina (IOAA; 0,125 mM; 0,2 mM; 0,25 mM) a rhodamin 6G (RH6G; 0,05 mM; 0,1 mM; 0,125 mM; 0,25 mM). Optimální koncentrace (0,4 mM IOA a 0,2 mM IOAA) byly stanoveny a aplikovány v asymetrické somatické hybridizaci. Testované koncentrace RH6G byly toxické. V kombinaci S. verrucosum (ozářené UV) + S. tuberosum cv. Karin (ošetřené 0,4 mM IAD) byla zaznamenána regenerace rostlin. (cs)
