  • Mitochondriální RNA-vážící proteiny MRP1 a MRP2 se vyskytují v heteromerickém komplexu, který patrně hraje roli v inserčním-delečním editování u trypanosom. Snížení hladiny mRNA pro MRP1 (gBP21) a-nebo MRP2 (gBP25) prostřednictvím interference RNA (RNAi) u procyklických stádií Trypanosoma brucei způsobuje závažnou poruchu růstu. Rovněž došlo ke ztrátě obou proteinů, což naznačuje jejich vzájemnou závislost ohledně stability. Vymizení MRP proteinů způsobilo významný pokles editovaných mRNA pro cyB a RPS12, ale pouze malé snížení editovaných cox2, cox3 a A6. Avšak množství editované mRNA pro ND7 se v dvojitých knock-downech 5x zvýšilo. Navíc způsobila eliminace MRP proteinů snížení hladiny mRNA pro cox1, ND4 a ND5, což jsou needitované molekuly. Tyto výsledky naznačují, že MRP1 a MRP2 nejsou pro samotné editování RNA nezbytné, avšak hrají regulační úlohu při editování specifických transkriptů a zúčastňují se dalších úprav RNA molekul. (cs)
  • Mitochondrial RNA binding proteins MRP1 and MRP2 occur in a heteromeric complex that appears to play a role in U-insertion/deletion editing in trypanosomes. Reduction in the levels of MRP 1 (gBP21) and/or MRP2 (gBP25) mRNA by RNA interference (RNAi) in procyclic Trypanosoma brucei resulted in severe growth inhibition. It also resulted in the loss of both proteins, even when only one of the MRPs mRNAs was reduced, indicating a mutual dependence for stability. Elimination of the MRPs gave rise to substantially reduced levels of edited cyB and RPS12 mRNAs, but little or no reduction of the level of edited cox2, cox3 and A6 mRNAs, as measured by poisoned primer extension analyses. In contrast, edited ND7 mRNA was increased five-fold in MRP1+2 double knockdown cells. These data indicate that MRP1 and MRP2 are not essential for RNA editing per se, but rather play a regulatory role in the editing of specific transcripts and other RNA processing activities.
