  • The relationship of flow cytometry data and epifluorescence microscopy measurements was assessed in bacterioplankton samples from 80 lakes to estimate bacterial biovolume and cell size distribution. Total counts of cells (abundances) estimated by both methods were significantly related. To explore the relationship between side scatter (SSC) or DAPI fluorescence data and cell volume (microscopy values), three classes with comparable number of observations were established for each parameter. SSC appeared to be the best cell volume predictor and significant relationships were observed between the frequencies of small classes of both parameters (r2 = 0.545, p< 0.001, n= 80), and in addition between the frequencies of the largest classes (r2 = 0.544, p< 0.001, n= 80). A reliable bacteria biomass prediction can be obtained from SSC frequency classes. FC can be used for a proper estimate of natural bacterioplankton biomass if sample heterogeneity is considered.

  • Vztah mezi údaji flow cytometrie a epifluorescenční mikroskopie (bakteriální buněčné objemy a jejich distribuce) byl sledován na vzorcích bakterioplanktonu z 80 jezer. Celkové počty (abundance) oběma metodami byly srovnatelné. Pro zhodnocení vztahu mezi „side scatter“ (SSC) a údaji o buněčných objemech z měření s použitím DAPI byly vzorky rozděleny do 3 tříd se srovnatelnou četností. S použitím SSC byly nejlépe odhadnuty buněčné objemy. Mezi frekvencemi v nejmenší a v největší třídě obou srovnávaných parametrů byly nalezeny signifikantní vztahy: (r2 = 0.545, p< 0.001, n= 80) a (r2 = 0.544, p< 0.001, n= 80). Pomocí tříd frekvencí SSC lze dobře odhadovat bakteriální biomasu. Flow cytometrii je možno použít k odhadu biomasy přírodního bakterioplanktonu, vezmeme-li v úvahu heterogenitu vzorku. (cs)
