  • Suitability of different methods (staining, concentration-sedimentation, concentration-floatation, combined floatation-sedimentation) for routine diagnostics of Cryptosporidium spp was compared. The methods were tested with two concentrations (1 x 105 and 1 x 106/g) of C. parvum and C. andersoni. The methods were evaluated using light microscope, magnification 400x for concentration methods and 1000x for stained samples respectively. Specificity of staining methods was 95-100%. Ziehl-Neelsen is more suitable for identification of C. andersoni and modified Miláček-Vítovec for identification of C. parvum. Concerning specificity and sensitivity, the floatation-concentration method by Sheather was found to provide the best results of all used methods. The merthiolate iodine formaldehyde concentration (MIFC) method was the least specific one. The least suitable method concerning sensitivity and costs was the floatation method with caesium chloride (CsCl) with a specificity of 29%.

  • Byla porovnána vhodnost různých rutinních metod (barvicí, sedimentační, flotační, flotačně-sedimentační) pro diagnostiku kryptosporidií. Metody by testovány s dvěmi koncentracemi C. parvum a C. andersoni ((1 x 105 a 1 x 106/g) a vyhodnoceny ve světelném mikroskopu při zvětšení 400x pro koncentrační a 1000x pro barvicí metody. Specifita barvicích metod se pohyboval mezi 95-100%. Metoda dle Ziehl-Nielsena byla vhodnější pro identifikaci C. andersoni a dle Miláčka-Vítovce pro oocysty C. parvum. Flotační metoda dle Sheathera byla vyhodnocena jako nejvhodnější z hlediska specifity a senzitivity. Naopak tzv. MIFC metoda byla jedna z nejhorších. Flotační metoda s použitím cesium chloridu se specifikou 29 % byla z hlediska senzitivity a finanční nákladnosti vyhodnocena jako méně vhodná. (cs)
