  • Bílkoviny představují důležitou část zrna ječmene, hlavně z pohledu potravinářských aplikací. V této práci byl hodnocen obsah celkového proteinu a relativní zastoupení proteinových frakcí dle rozpustnosti u 20 genotypů (odrůdy a novošlechtění) z celého světa. Použita byla modifikovaná metoda extrakce proteinových frakcí (albuminy + globuliny, prolaminy, gluteliny) založená na principu Osbornovy metody. Obsah celkového proteinu a proteinových frakcí byl určen jako obsah celkového dusíku v sušině (N x 6.25) modifikovanou metodou dle Dumase na elementárním analyzátoru FLASH EA1112 (ThermoQuest, Itálie).Genotypová variabilita mezi proteinovými frakcemi byla hodnocena elektroforézou (SDS-PAGE). U obsahu celkového proteinu byly nalezeny statisticky významnérozdíly mezi analyzovanými genotypy. Hodnoty obsahu celkového proteinu se pohybovaly v rozmezí od 9 do 21 % sušiny. Významné rozdíly byly také nalezeny u jednotlivých proteinových frakcí - průměrné proporce albuminů+globulinů, prolaminů a glutelinů předs (cs)
  • Proteins represent an important part of barley grain, mainly from the point of view of the food industry application. In this presentation, 20 barley genotypes (cultivars and new breeding lines) of world-wide range were evaluated for their total proteincontent and relative percentage of protein solubility fraction. The modified method of extraction of protein fractions (Albumins + Globulins, Prolamins, Glutelins) based on the Osborne's principle was used. Total protein content and protein fraction contents were determined as total nitrogen content in dry matter (N x 6.25) by modified Dumas method on elementar analyser Flash EA 1112 (ThermoQuest, Italy). Genotype variability in protein fraction composition was evaluated by electrophoresis (SDS-PAGE).In total protein content, the statistically significant differences among analyzed genotypes were found.
