  • Plant biotechnology, especially genetic transformation, represents one of powerful tools for the improvement of various important crops throughout the introduction of foreign genes into the plant genome. Although different transformation procedures have been developed so far, Agrobacterium tumefaciens-mediated transformation represents still one of the most efficient ways, with various desirable features. Efficient employment of marker genes allowed for critical assessments of various transformation and cultivation protocols, step by step. Since the agrobacterial transformations highly rely on the close adhesion of bacteria to wounded cell(s), procedures allowing for easier access of vector bacteria and subsequent transfer of T-DNA, could lead to a more efficient transformation. Various pretreatment and wounding procedures were found to be important factors leading to a more successful gene transfer and subsequently to increased transformation frequency. Procedures like treatment or wounding of plants tissues using various devices and tools, or sonication, were successfully employed in the transformation protocols of recalcitrant plant species indicating a possible way leading to the increased efficiency of targeted gene transfer.
