  • Využití bakterií Agrobacterium tumefaciens v transformačních systémech založených na jejich Ti plazmidech se stalo hlavním nástrojem pro inzerce cizích genů do rostlinných buněk, které posléze exprimují požadované genové produkty. Avšak u některých druhů, ke kterým náleží také len (Linum usitatissimum), dosáhly běžné transformační postupy poměrně omezených výsledků. V nedávné době bylo ukázáno, že sonikací zprostředkovaná agrobakteriální transformace (SAAT) výrazně zvyšuje úroveň transientní exprese reportérového genu GUS a frekvence transformací u některých rostliných druhů. Vzhledem k odlišnosti odezvy různých pletiv na SAAT, je třeba optimalizovat transformační protokoly pro jednotlivá pletiva. V této práci bylo využito SAAT ke zvýšení účinnosti transformace hypokotylových explantátů lnu. (cs)
  • The use of Agrobacterium tumefaciens in transformation system based on its Ti plazmid has become the major tool for insertion of foreign genes into plant cells which then express desirable gene products. However, in certain species, to which belongs also flax (Linum usitatissimum), the transformation methods has reached rather slow progress and mostly has lead to unsatisfactory results. Recently, it has been shown that sonication assisted Agrobacterium-mediated transformation (SAAT) greatly enhanced the levels of transient expression of a GUS reporter gene and transformation frequencies in some species. As tissues differ in their response to SAAT, each plant tissue needs the optimized protocol. Here, the SAAT was used to increase transformation efficiencies for flax hypocotyls.
