Description
| - Cílem práce bylo vyvinout markerovací systém založený na využití retrotransposonových sekvencí pro analýzu genetické diverzity kolekce lnu. Celkem bylo osekvenováno více než 100 kb a následně identifikováno celkem 27 odlišných retrotransposonových fragmentů. Následně bylo navrženo a testováno 75 PCR primerů z nichž 23 poskytly reprodukovatelný polymorfní profil. Tento systém byl testován na souboru 50 geograficky odlišných položkách lnu z kolekce Agritec, CR. Tyto výsledky poslouží k dlaší práci s cílem tvorby core kolekce lnu na pracovišti Agritec, ŠUmperk. - The goal of this study was to develop a molecular marker system for Linum based on retrotransposons and used it to germplasm diversity evaluation and flax core collection development. 75 PCR primers were designed and tested in IRAP (Inter Retrotransposon Amplified Polymorphism) protocol.We have identified 27 primers that displayed clear banding patterns and useful levels of polymorphism, yielding multiloci fingerprint. 23 primers have produced low multiloci pattern, but can be further combined. In summary, use of 3 to 4 single primer IRAP protocol have potential fully discriminate and identify tested set of 50 flax varieties. Based on primer used, 15 to 30 (100 to 3500 bp) PCR amplified fragments can be visualized, of which 5 to 25% was polymorphic eg. informative. All these results are used in further work aiming at core collection development from current over 2.000 accessions of flax collection hold at Agritec Ltd.
