  • V průběhu agrobakteriální transformace je kokultivace klíčovým krokem. Odrůdy polního hrachu Adept, Komet a Menhir byly použity v řadě experimentů s aplikací kokultivačních látek – acetosyringonu, L-cysteinu, dithiothreitolu, glutathionu, maceračních enzymů – celulázy a pektinázy. Byl také testován vliv režimu osvětlení – nepřetržité světlo nebo tma. Pozitivní vliv byl zaznamenán při aplikaci acetosyringonu a L-cysteinu. Glutathion neměl vliv na efektivitu transformace ani při opakování experimentů a při různých škálách koncentrací. Dithiotreitol a macerační enzymy, které byly použity každý zvlášť i v kombinaci, měly na efektivitu transformace negativní účinek. Režim nepřetržitého osvětlení měl pozitivní vliv na frekvenci explantátů s uniformní expresí GUS. (cs)
  • Agrobacterium-mediated genetic transformation includes several methodical step, the cocultivation step is a crucial period. Cultivars of dry-seed peas - Adept, Komet, Menhir were used in a set of experiments with application of various cocultivation substances, such as acetosyringone, L-cysteine, dithiothreitol, glutathione, macerating enzymes – cellulase and pectinase. The effect of continuous light and dark conditions during cocultivation were tested as well. Among all studied cocultivation substances, the positive effect was recorded only for acetosyringone and L-cysteine. Glutathione was without any positive or negative result. Dithiothreitol and both macerating enzymes had negative effect on frequency of GUS positive explants. Continous light during cocultivation possessed positive effect on the frequency of explants uniformly expressing GUS.
