  • Jeden z posledních trendů v oblasti rastrovací elektronové mikroskopie se zabývá pozorováním měkkých biologických vzorků v prostředí vysokého tlaku. Tento mikroskop je nazýván environmentální, nízko vakuový nebo mikroskop s proměnným tlakem. Pro pozorování biologických vzorků v jejich naturální podobě musí byt zachován specifický proces hydratace jež je výsledkem závislosti teploty na tlaku v prostoru komory vzorku ESEM. Dvě metody pro pozorování vlhkých biologických vzorků v jejich nezměněné, nebo pouzze málo změněné naturální podobě jsou popsány v tomto článku. (cs)
  • One of the recent trends in the field of scanning electron microscopy deals with observation of soft biological specimens at higher pressure in the specimen chamber. This microscopy is called environmental or low vacuum or variable pressure scanning electron microscopy. As wet biological specimens are to be observed in their natural state, specific process of hydration and temperature conditions in the dependence on pressure in the specimen chamber must be ensured. Two methods for observation of wet bbiological specimens in their unchanged or only a slightly changed natural state using the environmental scanning electron microscope are described in this article.
