  • Polyphenols exhibit significant antioxidant properties of wines. Their antioxidant capacity is commonly expressed as the equivalent weight of Trolox which is synthetic water-soluble analog of alfa-tocopherol (vitamin E), and is called %22Trolox equivalent antioxidant capacity%22 (TEAC). This contribution offers a direct determination of TEAC in real samples without the use of free radicals as it is in conventional spectrometric methods. An amperometric tyrosinase biosensor based on carbon paste electrode modified with carbon nanotubes (CPE/CNTs) is presented. The biosensor, CPE/CNTs/Tyrosinase/Nafion, was prepared by immobilization of tyrosinase enzyme together with Nafion on the surface of CPE/CNTs. All key experimental and instrumentals parameters have been investigated and optimal conditions were founded as follows: (i) amount of tyrosinase in Nafion membrane, 3.0 ?g; (ii) operating potential for amperometric detection, -0.25 V; (iii) speed of stirring, aprox. 400 rpm; (iv) supporting electrolyte, 0.1 mol L(-1) phosphate buffer solution, pH 7.0; (v) temperature 25 +-1 °C. Finally TEACs of wines were determined by hydrodynamic amperometry using the method of multiple standard additions.
