Description
  • The chapter provides overview of the theory and principles of asymmetric flow field flow fractionation (A4F). Effect of various operational parameters on the resolution of the A4F technique is discussed. For the sake of better understanding the A4F method is contrasted with widely known size exclusion chromatography (SEC). In contrast to SEC, in which the size restricted permeation of macromolecules into the pores of different size is the major separation mechanism, A4F separates the molecules without contribution of stationary phase. That is the most distinguished feature of the method that offers several advantages over SEC. The focus is on the combination of A4F with a multi-angle light scattering (MALS) detector that allows direct determination of molar mass and size distribution and characterization of branching. Theoretical and descriptive part is accomplished by several examples of experimental results obtained by the measurements of typical polymers, nanoparticles and proteins.
