  • Kyselina močová je konečným produktem metabolismu purinů u lidí. Ukázalo se, že kyselina močová je antioxidantem a reaguje s biologicky významnými oxidanty za vzniku alantoinu. Proto tedy byl alantoin navržen jako marker oxidačního stresu u lidí. Měřili jsme alantoin v plazmě pacientů s chronickým selháním ledvin před hemodialýzou (n=30), pacientů s non-insulin dependentním diabetem (n=30) a u dobrovolných dárců krve (n=30) pomocí metody založené na selektivní izolaci alantoinu ze vzorků deproteinované plazmy na AG 1-X8 pryskyřice a jeho derivatizaci na glyoxylát-2,4-dinitrofenylhydrazon. Při tomto stanovení neinterferují kyselina močová a glyoxylát. Separace glyoxylátu-2,4-dinitrofenylhydrazonu od ostatních hydrazonů byla dosažena pomocí HPLC v systému obrácenných fází s gradientovou elucí a UV/VIS detekcí při 360 nm. Analytické parametry této metody jsou dostačující s přesností v sérii CV 5,7%, mezi sériemi 8,3% a výtěžností 94,1%. Také byly stanoveny další parametry oxidačního stresu - malondialde (cs)
  • Uric acid is the end product of purine metabolism in humans. It has been pointed out, that uric acid acts as antioxidant and is capable to react with biologically relevant oxidants to form allantoin. Therefore, measurement of allantoin in humans was proposed as marker of oxidative stress. We estimated allantoin in human plasma obtained from the patients with chronic renal failure before hemodialysis (n = 30), the patients with non insulin dependent diabetes mellitus (n = 30) and blood donors (n = 30) using a method based on selective isolation of allantoin from deproteinized plasma samples on AG 1 X8 resin and its derivatization to glyoxylate 2,4 dinitrophenyl hydrazone. The method is free from urate and glyoxylate interferences. Separation of glyoxylate 2,4 dinitrophenyl hydrazone from other hydrazones was achieved on reversed phase HPLC with gradient elution and UV/VIS detection at 360 nm.
