  • Byly optimalizovány dvě metody. Jednak postup extrakce DNA dle Cenise (1992) a postup PCR dle Edwardse et al. (2001), jednak byl pro izolaci DNA a její následnou amplifikaci použit komerční set určený pro izolaci a amplifikaci genomické DNA z listů rostlin. Použití komerčního setu se pro detekci toxinogenních plísní rodu Fusarium jeví jako spolehlivější a rychlejší. Pro optimalizaci a stanovení citlivosti reakce byla použita sbírková kultura F. graminearum CCM F - 683. Selektivita metody byla ověřena na sbírkových kulturách plísní, které se mohou vyskytovat v reálných vzorcích potravin společně s plísněmi rodu Fusarium.Polymerasová řetězová reakce byla použita pro sledování přítomnosti Tri5 genu kódujícího tvorbu trichothecenových mykotoxinů. Bylo vyšetřeno 50 vzorků potravin a krmiv. Sedm z nich bylo pozitivních na přítomnost plísní rodu Fusarium. Pět druhů rodu Fusarium vyizolovaných ze vzorků obsahovalo v DNA Tri5 gen. U jednotlivých sbírkových plísní i plísní získaných ze vzorků byly rovněž sledov (cs)
  • Two methods of isolation of DNA were evaluated for successful identification of Fusarium species. The PCR was performed using primer specifically targeted to Tri5 gene. Extraction technique of DNA by Cenis (1992) and PCR procedure according to Edwards et al. (2001) was compared with DNA isolation and amplification using Plant PCR Kit. Sensitivity of the latter method was determined using reference strain of Fusarium graminearum CCM F-683 and selectivity was examined using reference cultures of different genera of fungi naturally occurred in food and feedstuff. Morphological features are important for species classification whereby PCR can distinguish between toxigenic and non-toxigenic fungi of Fusarium. These fungi could be detected within 2 days, while their identification by macroscopic and microscopic features takes 2-3 weeks. Thus the PCR technique has proved itself as reliable and rapid method for detection of toxigenic species of Fusarium.
