  • Byly sledovány problémy při imunomagnetické separaci listerií a byly porovnávány dva druhy imunomagnetických částic(Dynal BiotechxQuantum Magnetics). Pro zjištění nespecifických reakcí během IMS byly použity čisté kultury Listeria innocua, Arthrobacter spp., Bacillus subtilis, Citrobacter braakii, Escherichia coli, Enterobacter aerogenes, Enterobacter cloacae and Staphylococcus aureus. Ve většině případů vykazovaly nespecifické reakce gram pozitivní mikroorganismy, zejména St. aureus a Arthrobacter spp. Nespecifické reakce během IMS byly ověřeny na uměle inokulovaných potravinových vzorcích, ve kterých vykazovaly gram pozitivní bakterie nejvyšší procento záchytu. Byl rovněž studován vliv promývání pufry. (cs)
  • Problems occurring during the immunomagnetic separation (IMS) of Listeria using immunomagnetic particles Dynabeadsâ anti-Listeria (Dynal Biotech, Norway) were specified. Characteristics of these particles were compared with anti-Listeria spp. magnetite particles (Quantum Magnetics, USA). Pure cultures of Listeria innocua, Arthrobacter spp., Bacillus subtilis, Citrobacter braakii, Escherichia coli, Enterobacter aerogenes, Enterobacter cloacae and Staphylococcus aureus were used to evaluate non-specific reactions during IMS. Gram-positive microogranisms, especially Staphylococcus aureus and Arthrobacter spp. of the initial were found to be responsible for non-specific reactions in most cases. The capacity of Dynabeadsâ anti-Listeria particles was determined to be about 10 % pure cultures of Listeria spp., after 10 min of incubation.
